The Holy Sword weapon does not target enemies, you are experiencing a bug that can happen with many skills or attack (You can only actually target one enemy with an attack, you cannot select anything else.) If anyone experiences this bug the best thing I have found to do is to close the window and reboot the game, that one is currently out of my hands.

RPGMaker MV supports using the mouse to select spell/ability targets. If the mouse cursor happens to be over an enemy or player, keyboard target selection will fail.
